Most people who use peptides at home learn the process through trial and error. That means common mistakes repeat without the person noticing, and the result can be uneven absorption, more site pain, or lower-than-expected results. The good news is that identifying these mistakes is the first step to fixing them.

Needle angle and depth

One of the most frequent mistakes is using the wrong angle during insertion. For subcutaneous injection, which is the case for most peptides, the needle should enter at a 45 to 90 degree angle depending on your body composition. Using 90 degrees when you should be using 45 can cause the medication to reach the muscle instead of subcutaneous tissue, changing absorption.

Depth also matters. Shorter needles are recommended for people with less subcutaneous tissue. If you are using a longer needle than necessary, you might be injecting into the muscle. Not rotating injection sites

Injecting in the same spot every time causes lipodystrophy, which is the buildup of fat under the skin at that point. The tissue scars and absorption becomes irregular. The ideal approach is to rotate among at least three to five different body locations. The abdomen, thigh, and upper arm are the most common areas.

Aspirating before injecting

Aspiration — pulling back the plunger after inserting the needle to check for blood — is not necessary for subcutaneous injections. In intramuscular injections it can be a sign that you hit a vessel, but for subcutaneous tissue this step is unnecessary and can cause more tissue damage. If you have been aspirating before each subcutaneous injection, you can stop.

Injecting into irritated or bruised tissue

Injecting into an area that is already injured, bruised, or irritated compromises absorption and can cause more pain. Always choose a site that looks normal and is not tender. If you have injected in the same location several times in a row, give it time to recover before using it again.

Incorrect storage

Returning the peptide to the refrigerator immediately after injection is essential. Leaving the vial out of the fridge for hours compromises the stability of the formula. Freezing is also incorrect.
